東京都港区浜松町に位置する、都営地下鉄浅草線と大江戸線が乗り入れる駅になっております。 JR山手線・京浜東北線・東京モノレール羽田空港線の浜松町駅は乗換となります。両路線へは約100m離れています。

駅名の由来は、駅に近接している仏教寺院「増上寺」の総門を指す「大門」から由来します。増上寺へは三田線の芝公園駅の方が比較的近い。

浅草線ホームは相対式ホームで、エアポート快特以外の列車種別は浅草線内の各駅に停車します（泉岳寺～新橋駅間は全車両各駅停車）。 2019年（令和元年）11月中旬から、浅草線ホームでホームドアが設置されて稼働開始しました。浅草線内では新橋駅に次いで２駅目となります。2021年（令和３年）２月から副駅名「浜松町」を導入した（大江戸線は開業当初より導入済）。

大江戸線ホームは島式ホームであり、ホームが地下５階にあり、結構深い場所にあります。連絡通路・改札階は地下３階です。地上へ上がるエレベーターもあります。大江戸線のエレベーターは以前はKONE製でしたが、東芝に更新されました。改札外にある連絡通路には、大門の歴史を記載されたパネルも設置されている。

当駅は11ヶ所の出入口が存在し、日本生命浜松町クレアタワーへは、大江戸線側のＢ５出口で直結します。 港区役所へは、浅草線側のＡ６出口から徒歩５分程度で行けます。

１日の乗降客数も大規模であり、約11～14万人程度が利用するため、平日のラッシュ時間帯は混雑しますね。近くにはJR浜松町駅・モノレール浜松町駅があって乗換客も結構多い。浅草線内では押上・泉岳寺・三田駅に次いで第４の規模を、大江戸線内では新宿駅に次いで第２の規模を誇ります。
