Zgrada u kojoj se nalazi Muzej pozorišne umetnosti Srbije izgrađena je 1836.god kao kuća trgovca Miloja Božića. Nalazi se pored tadašnje Dositejeve Velike škole, a danas Muzeja Vuka i Dositeja. Muzej je osnovan 1901.god na inicijativu Branislava Nušića, a otvoren je 1950.god sa izložbom koju su činili eksponati sakupljeni tokom prethodnih 50 godina. U muzeju se nalaze predmeti koji prikazuju razvoj pozorišta u Srbiji od 13-tog veka do danas. Muzej je organizovan po raznim zbirkama: fotografije, pisma, rukopisi, plakati i programi,.. Postavka nije velika ali je zanimljiva.
